Oral administration of pharmaceutical active ingredients (API’s) is the preferred route for the majority of medical treatments. Advantages of oral delivery are the high compliance of the patient, the possibility of self-administration, relatively low costs of manufacturing and low invasiveness.
Orally administrated drugs encounter several physiological barriers, such as degradation by the acidic environment and enzymes in the gastrointestinal tract (GIT), metabolism by enzymes of luminal, brush border and cytosolic origin, and the permeability of the membrane. Prior to intestinal absorption, the drug must dissolve in the fluids of the GIT and by active or passive permeation pass through the small intestinal membrane.
Alternative ways of protecting and transporting API's through GIT are highly relevant, and in this current project, microcontainers are being explored in vitro, in situ and in vivo as vehicles for API's by oral drug delivery.
